探讨了采用寡聚环糊精键合新型凝胶层析介质从葛根黄酮中分离制备葛根素的新方法。介质为寡聚环糊精键合Sepharose HP,配基浓度为18 mg/ml湿凝胶。分离在440mm*35mm i. d.的色谱柱中进行。优化并最终确定制备型分离条件为:流动相7%醋酸水溶液,流速10ml/min,进样20ml 15mg/ml的葛根黄酮溶液(溶剂为5%的乙醇)。收集得到的葛根素组分经旋转蒸发浓缩后用水结晶、重结晶,母液循环套用。葛根素的总收率达到32.4%,纯度大于97%。

Abstract: Oligo-cyclodextrin coupled Sepharose HP media was used for the preparative separation of puerarin from pueraria flavone. The best preparative separation was performed with a 440mm*35mm i. d. gel column using the mobile phase of 7% acetic acid at flow rate of 10 ml/min after the loading of 300mg sample in 20 ml 5% ethanol. Puerarin fractions of the preparative chromatography were collected and evaporated to dry state. After the procedures of decoloring, crystallization and recrystallization, the purity of the puerarin is higher than 97% and the total recovery of procedures
