摘要: [目的] 考察杠柳苷元经大鼠肝微粒体体外代谢的特点,对其代谢产物进行定性研究。[方法] 建立肝微粒体温孵体系,采用超高效液相串联飞行时间质谱技术,结合保留时间、精确质量数和离子的碎片特征对孵育后的代谢产物进行结构鉴定。[结果] 杠柳苷元经肝微粒体温孵后,可以检测到除原型以外的4个I相代谢产物。[结论] 杠柳苷元经大鼠肝微粒体温孵后,主要的代谢途径为脱水、脱氢和甲基化,为其体内代谢的研究提供了基础。

Abstract: [Objective] The metabolites of periplogenin in rat liver microsomes were studied using UPLC-Q-TOF/MS. [Methods] After establish the rat liver microsomal incubation system, the metabolites of periplogenin in rat liver microsomes were detected by ultra performance liquid chromatography/quadrupole-time-of-flight mass spectrometry combine with retention time, exact mass number and fragmentation characteristics of the ions. [Result] Besides of the parent component, four metabolites were determined as phase I metabolites in the rat liver microsomal incubation. [Conclusion] The results indicated that the dehydration, dehydrogenation and methylation were the major phase I metabolic pathways of periplogenin in vitro. The present study provided the theoretical basis for metabolism in vivo.
