摘要: 目的 观察吸入不同时间异氟醚对发育期大鼠海马组织β-catenin的蛋白表达的影响。方法 出生后7天雄性SD大鼠32只随机分成4组:吸入空气的对照组(A组),吸入2%异氟醚2小时组(B组),吸入2%异氟醚4小时组(C组),以及吸入2%异氟醚6小时组(D组),每组幼鼠处理中均无缺氧及CO2蓄积发生。实验结束 6小时后,将上述各组幼鼠随机分为RT-PCR实验组和western blot实验组,分别取海马组织检测β-catenin蛋白表达。结果 发育期SD大鼠海马组织β-catenin mRNA的表达:与对照组相比,B组和D组有统计学差异(P<0.05),C组表达虽有上调,但无统计学意义;发育期SD大鼠海马组织β-catenin蛋白的表达:与对照组比,B组、C组和D组明显上调(P<0.05), 其中D组显著上调(P<0.01)。结论 吸入2%异氟醚处理会导致发育期大鼠海马组织β-catenin蛋白的表达上调。

Abstract: Objective To compare the effect of different time course of isoflurane on β-catenin expression in the hippocampus of developing rats. Method Thirty-two 7-day-old SD male rats were randomly divided into four groups: control group (A) inhalation of O2 instead of isoflurane ,inhalation of 2% isoflurane 2 hours group (B) , inhalation of 2% isoflurane 4 hours group (C) and inhalation of 2% isoflurane 6 hours group (D). Rats in every group were no hypoxia and CO2 accumulation occurred in the process. 6 hours after treatment rats in every group were killed and hippocampus were removed immediately . Results In the group of PCR, compared with control group, the level of β-catenin was increased markedly (P<0.05) only in groups B and D. While group C has raised, there was no statistical significance; In the group of western bolt, compared with control group, the expression of β-catenin in groups of B, C, D were raised markedly (P<0.05), in which the group D rised significantly (P<0.01). Conclusions Treatment with 2% isoflurane can result in upregulation of β-catenin expression in the hippocampus of developing rats.
