摘要: 以森林草莓(Fragaria vesca)为试材,利用RT-PCR技术克隆得到着丝粒特异性组蛋白H3变异体(CENH3)基因,并成功构建了草莓CENH3基因RNAi植物表达载体-pRNAi-CENH3,并将其导入根癌农杆菌EHA105中。通过果实注射农杆菌进行功能基因验证,结果显示注射了EHA105(pRNAi-CENH3)的草莓果实的生长发育都被不同程度地抑制。本研究初步表明CENH3基因的沉默对草莓细胞的分裂产生影响。

Abstract: Centromere-specific histone H3 variant (CENH3) gene was cloned from Fragaria vesca by RT-PCR. The RNAi vector for strawberry CENH3 gene was successfully constructed and introduced into Agrobacterium tumefaciens strain EHA105. Transient expression by fruit injection of Agrobacterium carrying plant expression vector was used to validate the gene function. It showed that the development of strawberry fruits injected with EHA105(pRNAi-CENH3) has been inhibited with varying degrees. In this study, the result preliminarily showed that the effect of silencing of strawberry CENH3 gene on the division of strawberry cells.
