摘要: 目的 观察内源性抗氧化剂牛磺酸对糖尿病大鼠早期肾损害的保护作用及肾皮质凝集素样氧化低密度脂蛋白受体-1(LOX-1)和细胞间粘附分子-1(ICAM-1)表达的影响,并初步探讨其可能的作用机制。方法 6周龄雄性Wistar大鼠随机取出10只作为正常对照组(NC组),其余给予STZ(60mg/kg,i.p)诱导1型糖尿病模型,成模后随机分为糖尿病组(DM组,n=10),糖尿病牛磺酸干预组(DM+Tau组,n=10,饮水中加入1%牛磺酸)。4周后处死大鼠,测定肾脏肥大指数(左肾重量/体重)、血糖及血清尿素氮(BUN)、肌酐(Cr)水平;留取肾脏组织测定肾组织丙二醛(MDA)、谷胱甘肽过氧物酶(GSH-Px)的水平作为氧化应激参数,定性定量检测肾皮质LOX-1、ICAM-1蛋白及基因的表达。结果 同糖尿病组相比,牛磺酸干预的糖尿病大鼠肾脏肥大指数、血清BUN、Cr、肾组织MDA水平明显降低(P均< 0.01),而肾组织GSH-Px水平明显升高(P<0.05);而肾脏过度表达的LOX-1和ICAM-1蛋白及基因水平明显降低(P均< 0.05)。两组比较差异有统计学意义。结论 牛磺酸对糖尿病大鼠早期肾损伤具有保护作用,这种保护作用与其抗氧化特性降调糖尿病大鼠肾组织LOX-1和ICAM-1的表达有关。

Abstract: Objective To investigate the renoprotective effects and possibel mechanisms involved LOX-1 and ICAM-1 expression of taurine in early renal injury. Methods 6-week-old male Wistar rats were divided into three groups: normal control (NC), diabetes mellitus (DM) and taurine-treated DM (DM+Tau) group. Diabetes was induced by a sigle injection of streptozotocin (STZ, 60mg/kg, i.p.). After onset of diabetes, drinking water containing 1% taurine were given to rats in the DM+TAU group. After 4 weeks treatment, Blood glucose (BG), serum levels of creatinine (sCr) and BUN, LOX-1 and ICAM-1 expression (protein and gene) in kidney cortices were estimated. Meanwhile, renal malondialdehyde (MDA) levels and glutathione peroxidase (GSH-Px) activities were examined as parameters of oxidative stress in diabetic rats. Results For DM+TAU rats, when compared with DM rats, the levels of serum BUN, sCr and renal MDA were reduced and the activities of renal GSH-Px were increased, but the BG levels were not influenced. Simultaneously, taurine reduced overexpression of LOX-1 and ICAM-1 on kidney cortices of diabetic rats. Conclusion Taurine showed protective effects against early renal injury in diabetic rats. These renoprotective effects may be caused by suppression of LOX-1 and subsequently ICAM-1 overexpression on kidney cortex via its antioxidative property.
