摘要: 目的:研究哮喘大鼠肺组织和外周血淋巴细胞中SUMF2的表达水平,为进一步探究哮喘的有效检测指标及靶位治疗奠定实验基础。方法:用OVA致敏激发大鼠哮喘模型,生理盐水和硫酸铝钾致敏作为对照组。酶联免疫吸附试验(ELISA)测定肺泡灌洗液(BALF)中IL-13和IL-4的浓度;HE染色观察气道炎症情况。实时定量聚合酶链式反应(Real-time PCR)方法检测各组大鼠肺组织和外周血淋巴细胞SUMF2及IL-13的mRNA表达情况;免疫组化测定肺组织SUMF2和IL-13的蛋白表达情况。应用SPSS17.0进行数据分析。结果:哮喘组肺组织中SUMF2蛋白的表达水平明显低于两对照组(P<0.05),IL-13蛋白的表达水平明显高于两对照组(P<0.05)。哮喘大鼠外周血淋巴细胞和肺组织中SUMF2的mRNA表达水平明显低于对照组(P<0.05),IL-13的mRNA表达水平明显高于对照组(P<0.05)。对照组之间SUMF2和IL-13的表达在mRNA和蛋白水平均无显著差异。结论:哮喘大鼠肺组织和外周血中均有SUMF2和IL-13的表达:SUMF2低表达,IL-13高表达。IL-13 的高表达参与了哮喘的病理生理过程,SUMF2的低表达可能成为哮喘的有效检测指标并可能成为哮喘治疗的新靶点。

Abstract: Objective: To investigate the expression of SUMF2 and IL-13 in asthma rat lung tissue and peripheral blood lymphocytes. Lay the foundation diagnostics of asthma in order to further explore the effective diagnostic indicators and targeted therapy. Methods: Using OVA trigger asthma model rats, physiological saline and aluminium potassium sulfate trigger as control group. Enzyme-linked immunosorbent assay (ELISA) determination the concentration of IL - 13 and IL - 4 of bronchial Alveolar Lavage Fluid (BALF), HE staining to observe airway inflammation. SUMF2 and IL-13 mRNA was detected in rat lung tissue and peripheral blood lymphocyte with real-time PCR.SUMF2 and IL-13 protein expression was detected in lung tissue with immunohistochemical.The statistical software SPSS17.0 was used to analyze the experimental data.Results: The expression of SUMF2 protein in lung tissue was significantly lower than the two control group, the expression of IL - 13 protein in lung tissue was significantly higher than the two control group(P<0.05). The expression of SUMF2 mRNA in lung tissue and sthma rat peripheral blood lymphocytes was significantly lower than the two control group, the expression of IL - 13 mRNA is significantly higher than the control group(P<0.05).The expression of IL - 13 and SUMF2 has no significant difference in the mRNA and protein level between the two control group.Conclusions:Both IL - 13 and SUMF2 were detected in lung tissue and peripheral blood ,there expression of SUMF2 is lower expression,and IL - 13 is high expression. High expression of IL - 13 is participate in the pathophysiological process of asthma.The lower expression of SUMF2 in asthma could be an effectively detect indicator and may become new target for the treatment of asthma.
