摘要: 目的:探讨马尾松树皮提取物(PMBE)对黑色素瘤A375细胞体外迁移及侵袭作用的影响,并初步探讨其作用机制。方法:不同浓度的马尾松树皮提取物(PMBE)作用于黑色素瘤A375细胞,采用MTT法检测PMBE对黑色素瘤A375细胞增殖的影响;划痕实验检测PMBE对黑色素瘤A375细胞迁移能力的影响;体外侵袭实验(Transwell小室)检测PMBE对黑色素瘤A375细胞侵袭能力的影响;Western-blot检测PMBE对A375细胞基质金属蛋白酶-9表达水平的影响。 结果:随着PMBE的浓度增加,A375细胞增殖能力明显受到抑制,IC50值为25±2.4 μg/mL。PMBE浓度为5μg/mL、15μg/mL 时,细胞划痕的闭合能力分别下降19%、55%,细胞侵袭能力下降45%、73%。结论:PMBE能有效抑制黑色素瘤增殖和转移,其作用机制可能与降低黑色素瘤细胞运动能力,并且PMBE有潜力开发为针对黑色素瘤患者的抗癌药物。

Abstract: Objective:This study aimed to investigate the inhibitory effects of Pinus massoniana bark extracts (PMBE) on tumor metastasis in melanoma cancer A375 cells in vitro. Methods:Effects of Pinus massoniana bark extract on the viability of melanoma cancer A375 cells were detected by MTT assay. Melanoma cancer cell metastasis after exposure to Pinus massoniana bark extract were quantified by wound healing and Transwell assays, respectively. The expression of MMM-9 protein were evaluated by western blot. Rusults: Ovarian cancer cells following PMBPs treatment showed a dramatic reduction in cell viability in a dose-dependent manner, the IC50 value of PMBPs was 25±2.4 μg/mL. After cells were treated with 5 and 15 μg/mL PMBE, the cell invasion potentials were also decreased by 19% and 55%, the cell metastatic potentials were also decreased by 45% and 73%, respectively. Conclusion: Our findings indicated that PMBE had potential to develop as an anti-metastatic cancer drug for melanoma cancer patients.
